Work Performed
Consequence of Error/Judgement
Errors in DNA Sequencing and genotyping may lead to the loss of data and/or material, and/or the generation of erroneous data. This will necessitate the repetition of the work, requiring additional
reagents and causing delay to the client, both adversely affecting the integrity of the facility. Failure to monitor instrument performance may lead to poor analyses, requiring repetition. Errors in monitoring consumables may lead to the temporary suspension of the facility s services. Work will be discussed with the SBC Research Manager on a regular basis, and reasons for failed experiments, etc. identified for the purpose of potentially revising relevant SOPs.
Supervision Received
Research assistant works under the direction of the SBC Research Manager.
Supervision Given
Not expected to supervise other staff, but may be required to instruct members of the Unit and others in the use of the automated DNA sequencers, associated software, and ancillary procedures.
Minimum Qualifications
Completion of a relevant technical program or a university degree in a relevant discipline and a minimum three years of related experience or an equivalent combination of education and experience.

Job Summary
In the UBC Sequencing and Bioinformatics Consortium (SBC), operates automated DNA Sequencers and ancillary apparatus to service various laboratories; advises clients regarding services available,
form of samples to be submitted, and protocols and procedures related to Sanger sequencing and genotyping; prepares reagents for sequencing and genotyping services, and performs any necessary
related procedures as required; analyses data and monitors instrument performance, maintaining equipment as necessary to ensure efficient operation: this may include troubleshooting and performing routine repairs; monitors consumption of reagents and other supplies. The ability to communicate effectively and interact positively with a wide variety of clients is essential.

Organizational Status
- Reports to the SBC Research Manager.
- Liaises with technical services of suppliers as necessary.
- Liaises with UBC laboratories and academic at other institutions as requested to supply services in automated DNA Sequencing and genotyping.
- Trains other users in sequencing and genotyping techniques; operation of the automated DNA sequencer and other relevant technological advancements.
Work Performed
- Provides DNA Sequencing service, including (if required) purification of DNA to be sequenced, performance of cycle sequencing steps and postreaction clean-up, and operation of automated DNA sequencer to provide UBC laboratories and academics at other institutions with Sanger sequence data output to server through customized LIMS.
- Provides genotyping service to provide UBC laboratories and academics at other institutions with fragment analysis results as requested by the client.
- Complies standard operating procedures (SOP s) for all protocols.
- Advises clients regarding services available, form of samples to be submitted for each service, and may provide training on sequencing/genotyping protocols and procedures.
- Liaises with clients regarding sequencing or genotyping results; interprets data; troubleshoots, and advises clients if experimental design revisions are recommended.
- Maintains records of Sanger sequencing, genotyping, related procedures performed, and data elucidated. Keeps records of failed analyses.
- Maintains DNA sequencer and ancillary apparatus, as necessary, to ensure efficient and reproducible operation: this may include troubleshooting, and performing routine repairs.
- Prepares reagents for Sanger sequencing, genotyping and associated procedures from commercially-supplied materials.
- Monitors performance of sequencer via regular use of standard sequencing templates and internal genotyping standards for quality control.
- Performs other analytical and/or preparative work as requested.
- Maintains records of all work performed and procedures used.
- Monitors supplies utilized in the operation of the DNA Sequencing and genotyping services, re-stocking as necessary.
- Keeps abreast of new advances in relevant areas by reading original literature and technical bulletins.
- Trains other users in the operation of the automated DNA sequencers, associated software packages, and ancillary
apparatus. - Performs such other services as may be required by the SBC Research Manager.

Preferred Qualifications
- Undergraduate degree in a relevant discipline or Graduation from a technical college or institute. B.Sc. or M.Sc. in biological sciences or a related field preferred.
- Previous lab experience is required.
- Knowledge of Sanger sequencing and genotyping technologies is necessary.
- Experience in the use of an automated DNA sequencer.
- Computer skills: familiarity with PC computers is essential. Familiarity with sequence analysis, Word, Excel would be advantageous.
